BEVS at PSC | Cell Line


Protein Sciences has developed a new patented S. frugiperda cell line (expresSF+®) with well-defined growth characteristics that grows in serum-free media. expresSF+® cells contain no pathogenic viruses or other microbes, have no known cancer genes or cancer-causing viruses, do not induce tumors in animals, are grown in serum-free medium and are derived from an insect that is non-biting.

This cell line expresSF+® is covered by U.S. patent number 6,103,526 and corresponding foreign patent applications.

Serum
Manufacturing recombinant proteins in traditional cell systems often requires the use of fetal bovine serum, leading to significant cost and safety concerns. In addition, serum can represent up to 80% of the raw material cost of fermentation and often is in short supply.

Protein Sciences' system is not subject to these limitations. The FDA has allowed these serum-free expresSF+® cells to be used for production of four candidate products for Phase I/II human clinical trials.

Stability
The expresSF+® cells can be passed continuously for at least 6 months and more than 50 passages without change in viability.

Virus Creation
expresSF+® cells maintain a high viability and are highly permissive to infection with baculovirus, resulting in high titer virus stocks (typically >107-108 pfu/ml).

These cells are useful both in initial scale up and in large-scale manufacturing. The expresSF+® cells increase significantly in size during infection, which occurs synchronously at high density, resulting in increased cell mass and protein expression per cell.

Other insect cells lines do not support efficient virus production and therefore alternative cells have to be used for preparation of virus stocks. expresSF+® cells are excellent for generating high virus titer stocks and a low multiplicity of infection (MOI) can be used during scale up and production.
